Just caught this on Rock Paper Shotgun:

Cities in Motion
http://www.rockpapershotgun.com/2010/08/19/you-can-be-slow-or-fast-cities-in-motion/#more-36471

Might be good.  As far as I know there hasn't been a 3D transport game yet, or a modern one for that matter.

I can't tell from the trailer how much or little infrastructure you can actually build, or whether you only handle the actual transportation.

Just to add something to ChalmersAbrams otherwise paculiar posts, I noticed that Gamespot has this game listed and it mentions some interesting things about the game.  Also they have some pretty cool screenshots that show rural areas and show some slopes, so roads aren't all flat.  From what I can gather, though, the game doesn't seem to allow for the player to develope their own roads and such, like in Transport Tycoon.  So the strategy in the game probably won't involve designing your own road networks in the most effecient and effective way, but will focus on the strategy of setting up routes on the preexisting roads in an atempt to best facilitate the ever changing passenger demands.
